Análise de microssatélites em genoma plastidial de espécies do gênero Spondias

A família Anacardiaceae possui 79 gêneros, com 875 espécies, distribuídas em regiões tropicais, subtropicais e temperado. Spondias é o gênero de maior importância da família contendo 20 taxa sendo 10 de ocorrência neotropical. O objetivo desse trabalho foi analisar a distribuição de sequências de microssatélites em genomas de cloroplastos de quatro espécies do gênero Spondias. Os genomas foram desenvolvidos usando sequenciamento de DNA de nova geração e estão disponíveis no NCBI (S. tuberosa – KU756562, S. bahiensis – KU756561, S. dulcis - MK405229 e S. mombin – KY828469). Sequências de microssatélites foram analisadas usando o software phobos, implementado no Genieous. Os resultados mostraram 70 loci distribuídos nos quatro genomas. Foi observado que os genomas de S. tuberosas e S. bahiensis foram os mais similares, enquanto os genomas de S. mombin e S. dulcis foram os mais divergentes. Em todos os genomas, a ordem dos loci foram bastante similares, não havendo grande diferenças para as regiões LSC, SSC e IRs. Entre os motivos, foram detectados somente di- e tri-nucleotídeos, sendo o genoma de S. mombin o que mais apresentou trinucleotídeos. Entre os motivos, o AT/TA foi o mais abundante e para tri-nucleotídeos foi detectado somente o motivo AAT. No presente trabalho conclui-se que os genomas das espécies de Spondias possui grande similaridade na distribuição e abundância de loci de SSR, sugerindo pouca diversificação para a evolução de microssatélites.
The Anacardiaceae family has 79 genera, with 875 species, distributed in tropical, subtropical and temperate regions. Spondias is the most important genus of the family containing 20 taxa being 10 of neotropical occurrence. The objective of this work was to analyze the distribution of microsatellite sequences in chloroplast genomes of four species of the genus Spondias. Genomes were grown using next generation DNA sequencing and are available from NCBI (S. tuberosa - KU756562, S. bahiensis - KU756561, S. dulcis - MK405229 and Spondias mombin - KY828469). Microsatellite sequences were analyzed using phobos software, implemented in Genieous. The results showed 70 loci distributed in the four genomes. It was observed that the genomes of S. tuberosas and S. bahiensis were the most similar, while the genomes of S. mombin and S. dulcis were the most divergent. In all genomes, the order of the loci were quite similar, with no major differences for the LSC, SSC and IRs regions. Among the motifs, only di- and tri-nucleotides were detected, with the genome of S. mombin presenting the most trinucleotides. Among the motifs, AT / TA was the most abundant and for tri-nucleotides only the AAT motif was detected. In the present work it is concluded that the genomes of Spondias species have great similarity in the distribution and abundance of SSR loci, suggesting little diversification for the evolution of microsatellites.
