INTRODUÇÃO: A utilização da enzima
L-asparaginase (L-asparagina amidohidrolase, L-Asnase) no tratamento da
leucemia linfoblástica aguda tem exibido resultados excelentes e bem
estabelecidos com enzimas de Escherichia
coli e Erwiniachrysanthemi. No entanto, é comum a
ocorrência de efeitos colaterais, como hipersensibilidade e imunogenicidade,
devido às diferenças moleculares entre a forma enzimática bacteriana e a enzima
endógena humana. Portanto, a busca por outros subtipos que facilitem a aplicação
e com menores efeitos adversos é uma demanda eminente. OBJETIVO: realizar uma prospecção tecnológica
sobre novas fontes de L-ASNases proveniente de bactérias no tratamento da
leucemia linfoblástica aguda. METODOLOGIA: análise de documentos de patentes
depositados no período entre 2017 e 2023 nas bases de dados do INPI (Instituto
Nacional de Propriedade Industrial do Brasil), The LENS, WIPO (World
Intellectual Property Organization) e EPO (European Patent Office) por meio de combinação “Bacteria AND L-asparaginase
AND acute lymphoblastic leukemia” resultou em 8192 patentes, com 5080 após a
exclusão dos documentos duplicados e apenas 28 foram elegíveis após a aplicação
dos critérios de inclusão. RESULTADOS: diversas espécies bacterianas foram
investigadas, incluindo espécies mesófilas e extremófilas. A pesquisa revelou
que a criação de mutantes de L-ASNase expressa maior eficácia terapêutica, ao
empregar técnicas como mutagênese dirigida, recombinação genômica e peguilação
de proteínas. CONCLUSÃO: múltiplas espécies bacterianas foram exploradas para
produção de L-Asnase, ocorrendo inovação notável em espécies particulares e
pioneiras na produção, com ênfase para a pesquisa com isolados de bactérias
termofílicas. A modificação genética aprimora a atividade e estabilidade das
enzimas, enquanto a tendência se concentra em melhorar enzimas existentes em
vez de descobrir novas, dada sua eficiência versátil em aplicações industriais,
médicas e ambientais.
INTRODUCTION: The use of the enzyme L-asparaginase
(L-asparagine amidohydrolase, L-Asnase) in the treatment of acute lymphoblastic
leukemia has excellent and well-established results with enzymes from Escherichia coli and Erwinia
chrysanthemi. However, side
effects such as hypersensitivity and immunogenicity are common due to molecular
differences between the bacterial enzyme form and the human endogenous enzyme.
Therefore, the search for other subtypes that facilitate application and with
fewer adverse effects is an imminent demand. OBJECTIVE: to carry out
technological exploration into new sources of L-ASNases from bacteria in the
treatment of acute lymphoblastic leukemia. METHODOLOGY: analysis of patent
documents deposited in the period between 2017 and 2023 in the databases of
INPI (Brazilian National Institute of Industrial Property), The LENS, WIPO
(World Intellectual Property Organization) and EPO (European Patent Office)
through combination “Bacteria AND L-asparaginase AND acute lymphoblastic
leukemia” proven in 8192 patents, with 5080 after exclusion of duplicate
documents and only 28 were eligible after applying the inclusion criteria.
RESULTS: several bacterial species were investigated, including mesophilic and
extremophilic species. Research has revealed that the creation of L-ASNase
mutants expresses greater therapeutic efficacy, when employing techniques such
as directed mutagenesis, genomic recombination, and protein manipulation.
CONCLUSION: multiple bacterial species were explored to produce L-Asnase, with
notable innovations occurring in particular species and pioneers in production,
with an emphasis on research with isolates of thermophilic bacteria. Genetic
modification improves the activity and stability of enzymes, while the trend
focuses on improving existing enzymes rather than discovering new ones, given
their applied efficiency in industrial, medical and environmental applications.
